A este respecto, demostramos (Santillo et al., 2007) que la SOD1 es liberada activamente de los sinaptosomas del cerebro de las ratas as\u00ed como de las c\u00e9lulas GH3 de la pituitaria de las ratas que representan un buen modelo para estudiar la liberaci\u00f3n inducible de SOD1 ya que poseen toda la maquinaria proteica neuronal involucrada en la exocitosis de la ves\u00edcula sin\u00e1ptica.<\/p>\n<p class=\"p__6\">Adem\u00e1s, en el intento de evaluar el posible papel que desempe\u00f1a la exportaci\u00f3n de SOD1, recientemente demostramos, en la l\u00ednea celular del neuroblastoma SK-N-BE, que esta enzima es capaz, gracias a la participaci\u00f3n del receptor muscar\u00ednico M1, de activar el ERK1\/2 y el AKT de forma dosis y dependiente del tiempo. Este efecto se redujo notablemente mediante el silenciamiento del receptor M1, as\u00ed como mediante el uso del antagonista de la pirenzepina M1 (Damiano et al., 2013).<\/p>\n<p class=\"p__7\">Sin embargo, se ha demostrado que el FGF-1 y la isoforma de 18 kDa del FGF-2 son secretados por una v\u00eda alternativa que se traslada directamente del citoplasma al espacio extracelular. An\u00e1logamente, tambi\u00e9n se ha informado de que la interleucina 1\u03b2 (IL-1\u03b2) es secretada por una v\u00eda de exportaci\u00f3n vesicular no cl\u00e1sica. Las prote\u00ednas solubles contienen cl\u00e1sicamente p\u00e9ptidos de se\u00f1al N-terminal que las dirigen al aparato de translocaci\u00f3n del ret\u00edculo endopl\u00e1smico (ER) (Walter et al., 1984).<\/p>\n<p class=\"p__8\">Adem\u00e1s, la secreci\u00f3n prote\u00ednica no cl\u00e1sica depende tanto de la energ\u00eda como de la temperatura y puede ser estimulada o inhibida por diversos tratamientos (Cleves, 1997; Hughes, 1999). En el modelo de rata transg\u00e9nica SOD1G93A ALS se despleg\u00f3 la respuesta prote\u00ednica y se ha observado apoptosis inducida por el estr\u00e9s del RE (Atkin et al., 2006); tambi\u00e9n se ha demostrado una respuesta prote\u00ednica desplegada, que incluye la inducci\u00f3n de quinasas sensoras de estr\u00e9s, chaperones y mediadores apopt\u00f3ticos, en las motoneuronas de la m\u00e9dula espinal de pacientes humanos con la forma espor\u00e1dica de ALS (sALS) que no est\u00e1 restringida a las mutaciones de SOD1 (Atkin et al., 2008).<\/p>\n<p class=\"p__12\">El SOD1 y otras prote\u00ednas est\u00e1n mal plegadas en los fALS y en los sALS, pero no est\u00e1 claro c\u00f3mo esto desencadena el estr\u00e9s de ER, la fragmentaci\u00f3n del aparato de Golgi, la interrupci\u00f3n del transporte axonal y la apoptosis. Cerca de 20% de los fALS es causado por las mutaciones del gen SOD1 (Neumann et al., 2006).
