Les premi\u00e8res preuves exp\u00e9rimentales que certaines lign\u00e9es cellulaires pourraient \u00eatre capables de s\u00e9cr\u00e9ter la superoxyde dismutase Cu,Zn remontent \u00e0 de nombreuses ann\u00e9es, lorsque nous avons, pour la premi\u00e8re fois, montr\u00e9 la s\u00e9cr\u00e9tion de cette prot\u00e9ine par des exp\u00e9riences r\u00e9alis\u00e9es dans des h\u00e9patocytes et des fibroblastes (Mondola et al., 1996), des cellules de neuroblastome SK-N-BE (Mondola et al., 1998 ; Gomes et al., 2007 ; Polazzi et al., 2013) et dans des cellules \u00e9pith\u00e9liales d\u00e9riv\u00e9es du thymus (Cimini et al., 2002).<\/p>\n<p class=\"p__4\">En outre, nous avons d\u00e9montr\u00e9 que dans les cellules de neuroblastome humain SK-N-BE, qui pr\u00e9sentent une plus grande sensibilit\u00e9 \u00e0 la cytotoxicit\u00e9 induite par la privation de glucose en raison d'une sensibilit\u00e9 accrue aux ROS (Shutt et al., 2010), l'exportation de SOD1 a lieu dans des conditions normales et est accrue \u00e0 la suite d'un stress oxydatif (Mondola et al., 1996, 1998). Cet effet a \u00e9t\u00e9 remarquablement r\u00e9duit par la mise au silence du r\u00e9cepteur M1 ainsi que par l'utilisation de la pirenz\u00e9pine, un antagoniste M1 (Damiano et al., 2013).<\/p>\n<p class=\"p__7\">Cependant, il a \u00e9t\u00e9 d\u00e9montr\u00e9 que le FGF-1 et l'isoforme de 18 kDa du FGF-2 sont s\u00e9cr\u00e9t\u00e9s par une voie alternative directement transloqu\u00e9e du cytoplasme dans l'espace extracellulaire. Pr\u00e8s de 20% de l'EVAA est caus\u00e9e par des mutations du g\u00e8ne SOD1 (Neumann et al., 2006). En effet, la majorit\u00e9 des mutants SOD1 conservent leur activit\u00e9 enzymatique, ce qui sugg\u00e8re l'apparition d'une fonction de gain d'activit\u00e9 toxique plut\u00f4t qu'une simple perte de fonction (Strong et al., 2005 ; Dion et al., 2009).<\/p>\n<p class=\"p__13\">(2005) ont d\u00e9montr\u00e9 une alt\u00e9ration de la s\u00e9cr\u00e9tion extracellulaire constitutive de la SOD1 mutante dans les cellules NSC-34 qui induit de fr\u00e9quentes inclusions cytoplasmiques et une insolubilit\u00e9 des prot\u00e9ines. Ces donn\u00e9es \u00e9tablissent un lien entre la s\u00e9cr\u00e9tion d\u00e9ficiente de la SOD1 mutante et les agr\u00e9gats prot\u00e9iques intracellulaires et la toxicit\u00e9 dans les cellules NSC-34. En outre, ces auteurs ont montr\u00e9 que dans un mod\u00e8le de SLA chez le rat transg\u00e9nique, la perfusion intraspinale chronique de wt-SOD1 exog\u00e8ne humain retardait de mani\u00e8re significative la progression de la maladie, ce qui sugg\u00e8re un nouveau r\u00f4le extracellulaire pour la SOD1 dans la SLA ; par cons\u00e9quent, la d\u00e9livrance extracellulaire de wt-SOD1 humain pourrait am\u00e9liorer la maladie clinique chez les rats transg\u00e9niques atteints de SLA, ce qui confirme le nouveau r\u00f4le extracellulaire de la SOD1 mutante et de la wt-SOD1 dans la pathogen\u00e8se et la th\u00e9rapie de la SLA, respectivement.<\/p>\n<p class=\"p__14\">En outre, chez des souris transg\u00e9niques, porteuses de mutations SOD1, des effets toxiques sur les motoneurones par activation de la microglie ont \u00e9t\u00e9 observ\u00e9s (Urushitani et al., 2006 ; Zhao et al., 2010).
